LIMS for pharma QC laboratories: specifications, results, OOS and certificates of analysis

The QLogix LIMS is a laboratory information management system that records every in-process, release, stability and raw material result against an approved specification, on instruments in calibration and reference standards in date. The system calculates each result from the determinations as read and judges it against the limits when it is saved, so an out-of-specification or out-of-trend result is identified at entry, not at review. An out-of-specification result stands until an investigation linked to the sample finds an assignable cause; a passing repeat never replaces it. A second person who recorded no result reviews the sample, QA approves it, and the certificate of analysis is produced from the approved record with the signatures.

What the module does

  • Specifications per material and testing stage (in-process control, release, stability, raw material acceptance, investigation or retest), with each test's method, unit, reported decimals, determinations, limits, alert limits, maximum %RSD and limit of quantitation; tests drafted by the AI from the registered specification, monograph or method are marked as such and checked one by one.
  • Specification approval with electronic signatures and revision as a new version: the current version stays in force until the next is approved, and samples keep the version they were registered under.
  • Samples registered against the specification in force, with lot, sampling point, quantity, storage, due date and priority; stability samples carry the study, condition and time point; results are recorded only on a sample whose receipt is on the chain of custody.
  • Results entered as read under the analyst's own account with the system time; the analysis time checked, a late entry justified, and raw data files (chromatograms, printouts) fingerprinted with SHA-256 and linked to each result.
  • Calculation in exact decimal arithmetic with rounding half up to the reported decimals (USP General Notices 7.20) before the comparison with the limits; %RSD, individual determinations and values below the limit of quantitation handled by rule; out-of-trend results need a written evaluation.
  • Instrument control: calibration and qualification on each instrument, a result refused on an instrument out of service or out of calibration on the day of analysis, and a failed calibration that lists every sample tested on it since its last passing calibration.
  • Reference standards and reagents in quarantine until their certificate is checked and they are released; a result is refused on a standard that was not released or was past its expiry or retest date.
  • OOS management: an out-of-specification investigation opened from the sample with the result, determinations, instruments, standards and analyst filled in; invalidation only by that investigation's assignable cause; repeats with a reason; corrections only by the analyst who recorded the result, never into compliance.
  • Laboratory indicators, result trends as control charts with Nelson run rules and capability, CSV downloads and a results API; controlled interfaces for chromatography data systems, instrument software and the ERP, each validated before it is made active.

How it fits the platform

Investigations are built in: an OOS investigation opens from the sample, and a failed calibration opens a deviation investigation pre-filled with the instrument and every sample to assess. Results entered in the electronic lab notebook are posted to the LIMS sample with the run's reference, so the reviewer opens the work behind each result. A LIMS instrument can be linked to its calibration record and its maintenance asset in the CMMS, and a result is then accepted on it only while those records also say it is fit for use.

The MES registers in-process and release samples of a lot in the LIMS against the specification in force, and the batch is not dispositioned until release testing is approved. An instrument data system sends results with their raw data and reads the worklist; an ERP requests samples for a lot and reads the dispositions. A result received through an interface is checked exactly like a typed one and shows where it came from.

Where people decide

The AI drafts the tests from the specification document; the analyst checks them and the specification is approved with signatures. The analyst records what was read; the system calculates, judges and refuses what does not comply. A reviewer who recorded no result on the sample checks it against the raw data, and QA approves it with an electronic signature, at which point the system sets the conclusion and the certificate of analysis can be issued.

Questions about LIMS

What is a LIMS for pharma?

A LIMS (laboratory information management system) for pharma holds the QC laboratory's specifications, samples, results and approvals as GMP records. The QLogix LIMS records each result against an approved specification, checks the instruments and reference standards used at the time of analysis, judges the result, and routes the sample for second-person review and QA approval with electronic signatures.

How does the LIMS handle out-of-specification (OOS) results?

The result is judged out of specification when it is saved and shown in red on the sample. The OOS investigation opens from the sample with the result, determinations, instruments, standards and analyst filled in, linked both ways. Only an assignable cause found by that investigation invalidates the result, which stays on the record struck through, and a sample with an OOS result standing cannot be cancelled.

Can the LIMS issue a certificate of analysis?

Yes, once QA has approved the sample. The certificate of analysis is produced from the approved record with the signatures and the conclusion the system set from the results; before approval only a draft is available. Every export is recorded in the audit trail.

Can instruments and chromatography data systems send results?

Yes, through a controlled interface that is registered, bound to an API key, validated and then made active. An instrument data system reports only for the instruments assigned to it, every message is logged as accepted, rejected or duplicate, and a received result goes through the same calibration, standard, late entry and OOS checks as a typed one.

Is the audit trail Part 11 ready?

The LIMS is built to 21 CFR Part 11 and EU GMP Annex 11: each result is attributable to the signed-in analyst, reviewers and approvers sign with password re-entry, nothing is deleted, and every change is kept in a hash-chained audit trail. The site validates it for its intended use.

How is it licensed?

Per user per month, on its own or with the electronic lab notebook and other modules, ordered by purchase order from the client portal. There is no card payment or self-serve plan.
